Introduction
This gene encodes a cell surface protein that is a member of the immunoglobulin superfamily. By fluorescence in situ hybridization (FISH), Meyerhardt et al. (1997) localized NGN in 15q22, a region infrequently affected by alterations in cancer. Vielmetter et al. (1997) mapped NEO1 to 15q22.3-q23 by FISH. The encoded protein consists of four N-terminal immunoglobulin-like domains, six fibronectin type III domains, a transmembrane domain and a C-terminal internal domain that shares homology with the tumor suppressor candidate gene DCC. Rajagopalan et al. (2004) presented evidence that neogenin functions as a receptor for repulsive guidance molecule, or RGM (RGMA; 607362). Like Rgm, neogenin was expressed in a gradient across the embryonic chicken retina. The avoidance of Rgm by chicken temporal retinal axons was blocked by anti-neogenin antibody and the soluble neogenin ectodomain. Dorsal root ganglion axons were unresponsive to Rgm, but were converted to a responsive state by neogenin expression.

Function
Multi-functional cell surface receptor regulating cell adhesion in many diverse developmental processes, including neural tube and mammary gland formation, myogenesis and angiogenesis. Receptor for members of the BMP, netrin, and repulsive guidance molecule (RGM) families. Netrin-Neogenin interactions result in a chemoattractive axon guidance response and cell-cell adhesion, the interaction between NEO1/Neogenin and RGMa and RGMb induces a chemorepulsive response.
